Facilities

HPLC-PDA-CAD 

The Dionex 3000 system is used for standard HPLC separations. The connected Corona Veo RS aerosol discharge detector is a universal detector that provides detection of metabolites that have no chromophores or ionize poorly in the MS.

Technical features:

  • Binary pump with degasser unit
  • Temperature controlled automatic injector unit (fitting 120 1-ml sample vials)
  • Temperature controlled column compartment
  • Photodiode array (PDA) detector (UV-VIS range, 200-600 nm)
  • Corona Veo RS Charged Aerosol detector (CAD, Thermo Scientific)
  • Chromeleon Chromatography Data Systems software

Examples: Targeted analyses of plant metabolites from various plant materials such as glucosinolates or carbohydrates and amino acids

LC-QToFMS 

The Bruker Impact HD provides high mass resolution and high sensitivity. Those features allow measurements of accurate masses for even trace compounds and the determination of their sum formulas. Another important feature of the QToFMS is the possibility of conducting MS/MS experiments to elucidate the structures of unknown compounds.

Examples: 

  1. Analysis of plant defensive compounds controlling feeding and oviposition preferences in insects
  2. Analysis steroid glycosides involved in plant resistance
  3. Metabolomic profiling to study effects of root parasitism and shoot herbivory

LC-QqQMS

The Bruker EVOQ LC system can be used for quantification of known target analytes. It provides high sensitivity and robust quantification over a wide concentration range.

Technical features:

  • ESI and APCI source
  • Triple Quad Mass Spectrometer
  • Single Ion Monitoring; Multi Reaction Monitoring

Examples: 

  1. Quantification of phytohormones involved in plant development and plant growth (JA, ABA, IAA, SA, JA-ILE)
  2. Quantification of glucosinolates from shoots and roots of different Brassica species

GC-QqQMS

The Bruker SCION TQ can be used for the analysis of volatiles from plants, fungi or bacteria from trace level to high concentrations. In addition derivatized primary metabolites, like sugars, can be measured as well. The VOCs can be quantified and the mass spectrometer allows us to elucidate their chemical structure. The connected thermodesorption unit allows us to sample volatiles directly in the greenhouse or the field and to analyze them without further extraction. The instrument is also equipped with FID detector.

  • Thermodesorption Unit UNITY 2 (Markes) including 100 sample Autosampler ULTRA (Markes) and recollection option

  1. Analysis of herbivore induced plant volatiles by thermodesorption
  2. SPME analysis of volatiles emitted by growth promoting microbes,
  3. Analysis of different sugars from derivatized samples
